Yes, you would route the audio into an USB (or firewire) audio interface. The USB out of the Blofeld contains no sound, only MIDI data (Notes to play, sound parameters). I have a desktop blofeld, I've never taken it out on a gig or anything like that so I can't speak to how it'll handle that, I use it for recording into my computer or just jam with it a bit.
