To get full playback controls in Presentation mode in Keynote, all you need to do is make the placed audio icon larger, then move your mouse into that larger region while in presentation playback mode. The controller is not visible while editing your presentation, so yust make it large square and then check in presentation view.

If you play a video in keynote, you get controllers under it. But if you play audio there are no controllers. I think that's bad. The only thing I can think of is to make a video with a still image (for example the image of the musical instrument) and with the sound you want to play. Then you can use that video in stead of your audio file.

To access the audio playback controls while in presentation mode, just move your mouse into the region of the large icon (up to 400 pixel), and the controller will appear. You can start and stop sound playback at any time while the slide is showing. When you advance to the next slide, the sound playback automatically stops.
